- The character of Spider-Man was created by writer Stan Lee and artist Steve Ditko. He first appeared in Amazing Fantasy #15 in August, 1962 in a story titled "Spider-Man!" The character has since gone on to become one of the most well-known and enduring super-heroes of all time, having appeared in numerous comic book titles, as well as feature films, animated projects and video games.

- Welcome to The Amazing Spider-Man, Roy Thomas! Hope you survive the experience! This is the first issue of the series with Roy Thomas as writer. It is also the first issue of the series written by someone other than Stan Lee. Lee departed the title with Amazing Spider-Man #100, though he is still an editor on the series.
- This is the final issue of the series with a .15 cents cover price. Beginning with issue #102, the cover price raises to .25 cents per copy.
- This is the first appearance of Michael Morbius, aka Morbius, the Living Vampire. He has all of the characteristics of a traditional vampire except that he is not undead, thus the "Living Vampire" moniker. He is a vampire created through science. He appears next in Amazing Spider-Man #102.
- Peter Parker attempted to cure himself of having super-powers by way of an experimental serum in Amazing Spider-Man #100. Instead, the plan backfired and it gave him six arms.
- The plot device of Spider-Man with multiple sets of arms was also utilized in the "Morbius" episode of Spider-Man: The Animated Series.
